Rabeprazole
Rabeprazole is an antiulcer drug and a member of benzimidazoles and pyridines. It is also a Proton pump inhibitor. It is white and crystalline in nature. Rabeprazole is very solluble in water and methanol, freely solluble in alcohol, chloroform and ethyl acetate, insolluble in ether and n-hexane. The moleculer weight of rabeprazole is 359.4 g/mol and melting point is 99-100⁰c. It is also known as Aciphex,Pariets,Sodium Rabeprazole, Rabelon.

■Mechanism of action:-
It is an antisecratory compound and having histamine H2 receptor antagonist properties. It inhibits the gastric H+/K+/ATPase at the surface of the parietal cell, due to this its blocks the final step of gastric acid secretion. Rabeprazole chemically activated at pH1.2 with a half life of 78 seconds.
■Uses:-
Rabeprazole is used in the treatment of,
- Duodenal ulcer.
- Gastric ulcer.
- Gastroesophageal reflux disease.
- Zollinger-ellison syndromes.
- Helicobactor pylori infections
■Dosage:-
For duodenal ulcer the usual adult dose is 20mg orally per day for 4 weeks after morning meal.
For gastroesophageal reflux disease the usual adult dose is 20mg orally once a day for 4-8 weeks. The maintainance dose is 20mg orally per day for upto 2 months.
For the treatment of Zollinger-ellison syndromes the usual initial dose is 60mg orally per day for 4-8 weeks.
■Side-effects:-
- Bloating and swelling face,arms,hands or feet.
- Tiredness and weekness.
- Rapid weight gain.
- Unusual weight loss.
- Yellow eyes or skin.
- Tingling of the hands or feet.
