Mechanism of action of Remdesivir in the treatment of Covid-19
The Covid-19 virus contains s-proteins, these s-proteins bind with the AcE-2 receptor of the host(Human) membrane. Due to this, the viral RNA enters the host cell. RNA contains information in terms of sequences. This information allows the RNA to make RNA-dependent RNA polymerase(RdRp) proteins. These proteins play an important role in viral RNA replication.
Remdesivir is a prodrug. The inactive form of remdesivir is known as, GS-5734. When it is injected into our body by IV route, it is converted into GS-704277 with the help of enzyme esterase (CES). GS-704277 is further converted into GS-441525 with the help of a phosphor amidase enzyme. This is a almost active form of remdesivir. GS-441525 is converted into nucleoside triphosphate with the help of the nucleoside phosphate kinase enzyme. This is the fully active form of remdesivir. This nucleoside triphosphate contains Exo-nuclease, which prevents RNA replication by blocking the formation of RNA-dependent RNA polymerase proteins.
Remdesivir:-
It is an antiviral drug, which is used in the treatment of ebola virus infection, hepatitis-c, etc. It is a prodrug the inactive form of remdesivir is known as, GS-5734. Now a day it is used in the treatment of novel coronavirus (Covid-19). It is a delayed chain response inhibitor means, once it is administrated, not directly block the production of RNA, it takes some time. After three nucleotide addition, this drug inhibits RNA replication.
Structure:
The molecular formula of remdesivir is,[C27H35N6O8P].
The IUPAC name of remdesivir is, 2-ethylbutyl (2S)-2-[[[(2R,3S,4R,5R)-5-(4-aminopyrrolo[2,1-f][1,2,4]triazin-7-yl)-5-cyano-3,4-dihydroxyoxolan-2-yl]methoxy-phenoxyphosphoryl] amino] propanoate.
